Abstract [eng] In this work, a new method was described to apply magnesium pyrophosphate and DNA particles as a matrix for in vitro protein synthesis in microfluidic droplets. Single DNA molecules were encapsulated and amplified in 4 pl droplets. Optimized DNA amplification conditions were established to synthetize ~1 µm, magnesium pyrophosphate and DNA containing particles from single molecules. Also, reaction conditions were adjusted to reduce DNA molecule loss due to nonspecific adsorption on surfaces. Obtained particles were separated from reaction mixture. Finally, these particles were used to synthetize green fluorescent protein and β-galactosidase in microtiter plate and microfluidic droplets.
