Abstract [eng] The aim of the study was to identify periodontitis-related miRNAs and to evaluate the diagnostic potential of target miRNAs for periodontitis diagnostics in saliva, gingival crevicular fluid, and blood plasma, and to assess miRNA associations with rheumatoid arthritis and clinical-pathological parameters of both diseases. The dissertation consists of an introduction, literature review, materials and methods, results, conclusions, bibliography, summary and author's list of publications. A genome-wide miRNA profiling which enables the evaluation of 2569 miRNAs was performed in gingival tissues and four potentially periodontitis-associated miRNAs were selected: miR-140-3p, miR-145-5p, miR-146a-5p and miR-195-5p. The associations between periodontitis and rheumatoid arthritis clinical course which was observed in the study sample was also revealed through alterations in miRNA expression profile in gingival tissue. Assessing miR-140-3p, miR-145-5p, miR-146a-5p and miR-195-5p quantity in gingival crevicular fluid, saliva, and blood plasma it was revealed that miRNAs detected in gingival crevicular fluid had better diagnostic value for periodontitis diagnostics as compared to miRNAs detected in saliva or blood plasma. Also, combination of miRNAs presented with better diagnostic properties as compared to individual miRNAs.
