Abstract [eng] Matrix solid-phase dispersion (MSPD) and ultra-performance liquid chromatography (UPLC) techniques were developed for the rapid and effective determination of organic compounds in solid matrixes. For this purpose two analyte/sample matrix systems were selected: carbadox and olaquindox in animal feed and Sudan dyes in chili powder. The separation of carbadox and olaquindox by UPLC under hydrophilic interaction chromatographis conditions was investigated. Using this technique the separation time was 10-times shortened and about 3-4-times higher efficiency with better resolution was achieved compared to conventional reversed-phase chromatography. Three solid phases as solid supports for MSPD of carbadox and olaquindox from feeds were compared and the cleanest extracts were obtained with octadecylsilica. Protonic solvents (methanol and ethanol) exhibited higher eluting strength, whereas acetonitrile was more selective. In the second part of the dissertation UPLC conditions for the separation of four Sudan dyes were optimized. MSPD conditions for the extraction of dyes from chili powders were studied. MSPD-UPLC method was applied in the analysis of real samples. The MSPD-UPLC technique allows extraction and clean-up to be carried out into a single step, without additional purification of the extracts, is very simple and significantly faster compared to existing techniques.
