Rapid population aging and the high burden of preventable age related diseases make it necessary to search for solutions that could extend healthy lifespan. Dietary supplements targeting mechanisms of aging are widely used. However, clinical evidence demonstrating their efficacy and safety remains limited. This thesis analyzed the effect of the TLM01 dietary supplement combination in healthy adults seeking to slow aging processes. TLM01 consisted of NAD⁺ 500 mg, resveratrol 1000 mg, berberine 1000 mg, quercetin 1000 mg, and fisetin 500 mg. The aim of the study was to evaluate whether three months of supplementation has a health promoting effect in individuals who use dietary supplements according to the intended recommendations and whether it may positively affect aging related biomarkers and indicate an effect on aging processes. This thesis analyzed data from an open label comparative group study. The study included 99 participants (79 used TLM01, while 20 formed a non supplemented control group). Blood and saliva samples were collected at baseline and after three months. In total, 20 biomarkers and the participants’ epigenetic age were evaluated. Data were analyzed at three levels: classical biostatistics for changes in individual biomarkers, multivariate systems biology analysis for the overall biomarker profile response, and machine learning and intervention-response methods oriented toward personalized medicine. The analysis did not show that TLM01 slowed biological aging over three months. The primary endpoint, biological age, did not improve and, after adjustment for baseline group differences, was unfavorable for the TLM01 group. NAD⁺ concentration increased in both groups, so this change cannot be attributed solely to supplement use. The most consistent favorable signal was related to the lipid profile, particularly LDL and non-HDL-C. However, after adjustment for baseline group differences, this effect was not statistically significant. In summary, TLM01 did not demonstrate a clear biological aging slowing effect, but the lipid changes and differences in individual response support the need for further research. |
