The mechanism of spacer capture by type II CRISPR-Cas systems remains enigmatic. Wilkinson et al. report cryo-EM structures of three macromolecular assemblies of the Cas1, Cas2, and Csn2 proteins. All three complexes contain duplex DNA running along a central channel. A filamentous form contains a continuous duplex and smaller units contain shorter sections of ∼30 bp. |
