Abstract [eng] |
Objective. Simulating human heart infarction in an animal (rabbit) organism, to implant autologous myogenic stem cells into the pathological focus of the animal heart and to confirm stem cell existence in the tissue; to evaluate the safety of cell therapy. Methods. Heart operation on laboratory animals (rabbits); primary myogenic cell lines – origin and maintenance in the culture in vitro; labeling stem cells using vital dyes or genetic manipulations; cell transplantation into animal heart; histological evaluation of the tissue. Results. In the rabbit organism, an experimental model of heart infarction was acquired; the heart infarction was confirmed histologically; the labeling of stem cells by using vital dyes and genetic manipulations was applied in vitro; histological existence of stem cells was confirmed in heart tissue. Conclusions. In laboratory animals (rabbits), an experimental model system of cellular cardiomyoplasty was acquired. The model is intended to study the possibilities of cellular therapy in clinical practice. The study of cell transplantation into and integration in the pathological focus confirmed the possibilities of heart regeneration by using stem cell therapy. |
