Abstract [eng] Development of Controlled Gene Expression System for Thermophylic Geobacillus and Parageobacillus spp. bacteria Members of the genus Geobacillus and Parageobacillus bacteria have unique metabolic properties and are able to synthesise high variety of chemical compounds and thermostable enzymes. The ability to modify their genomes, introduce new genes and overexpress existing genes would increase their usage in biotechnology. In this study, we were creating a new gene expression system called pMS using the autoinduction mechanism of bacteriocin geobacilin I found in Geobacillus thermodenitrificans bacteria. We inserted PgeoA, TgeoA, geoR and GeoK genes used in autoinduction mechanism of geobacilin I into pUCG18 plasmid and created pMS vector. Subsequently, we inserted superfolder green fluorescent protein gene between PgeoA and TgeoA genes, to be able to determine the efficiency of the system. Unfortunately, we weren’t able to insert pMS-GFP vector into geobacili. In order to expand the range of possible hosts, we were changing resistance to kanamycin gene in pMS vector to resistance to chloramphenicol gene.
