Abstract [eng] NF-kB factors are broadly expressed in many different cells, they are important in many cellular processes. NF-kB pro-inflammatory signaling pathway stimulates expression of hundreds of genes and it has been implicated in many diseases. NF-kB reporter systems can be used for different purposes, for example to analyse response to the LPS stimulation. The aim of this study was to design NF-kB reporter systems and analyze their efficiency using HEK293T and A549 cell lines. These reporter systems were designed based on CCL2 and ELAM promoters. Synthetic NF-kB binding sites, GFP reporter gene were also used in the design of reporter systems. It is known that LPS stimulates NF-kB activation, thus LPS was used as an activator to promote GFP expression. It was established that HEK293T cells showed slightly higher GFP expression without LPS stimulation. However, A549 cell line showed slightly higher GFP expression when stimulated with LPS. The study also revealed that pAcGFP-5xNF-kB-CCL2-2 reporter system was slightly more effective. Taken together, NF-kB reporter systems did not show a significant response to LPS stimulation.
