Abstract [eng] Breast cancer – one of the most common cancer forms, and it is mostly diagnosed among women. Although there are some therapies designed against some forms of breast cancer, developed resistance to drugs and tumour renewal are big issues. It was observed, that extracellular matrix around a tumour becomes stiffer comparing to a healthy tissue nearby. In this way, tomour formation creates favourable conditions, which let cancerous cells to develop invasive and metastatic phenotypes. Because of cells’ sensitivity to mechanical signals, level of surounding environment’s stiffness influences cell shape, motility and formation of stem cell-likephenotypes. Polyacrylamide gels – useful tool to create different stiffness enviromenments, which may mimic physical properties of healthy and cancerous tissue for in vitro cell studies. c-Jun – transcription factor, which is regulated by MAPK signal cascades and possibly participates in cell’s migration and proliferation responses dependent on environment stiffness, together with transcription cofactors YAP/TAZ. This study’s goal was to compare morphology and behavior differences, depending on different environment stiffness and different expression level of c-Jun.
