Tau Protein (2N4R Isoform) Synthesis and Purification Alzheimer's disease is the most common and best-studied disease caused by amyloid proteins, it is characterized by amyloid plaques and neurofibrillary tangles in the brain, with associated loss of synapses and neurons, resulting in cognitive deficits and dementia. Tau protein is the main component of neurofibrillary tangles. Up to now, there are no drugs to treat this disease. Research of the potential inhibitors of the disorder requires large amounts of protein. The aim of this thesis was to synthesize and purify the Tau protein 2N4R isoform fused with SUMO-His tag. The expression of the gene encoding the Tau protein 2NR4 isoform fused with SUMO-His-tag was successfully obtained in E. coli BL21(DE3) bacterial cells. The best gene expression was found by culturing bacteria at 37 °C temperature and inducing with 0,6 mM IPTG at 0,7 optical density. 5,4 L of E. coli bacterial culture was grown and 22,6 g of biomass was obtained. Due to the declared COVID-19 pandemic, protein purification studies have been not performed, but the expected results have been described. |
